● Please unpack the instrument carefully and
confirm that all components have been
successfully delivered without damage that may
have occurred during transportation

● The following components are included in the
delivery of the TissueGrinder:
1× TissueGrinder
1x USB cord
1× Power cord
1x Power Adapter
1× USB Stick
1× Quick Installation Guide
● Carefully remove the device from
the cardboard box
● Place the instrument on a secure
and stable workbench
● Connect the power adapter to the
power grid and to the instrument
● Connect the USB cable to the
instrument and to the PC
● Set the switch located above the
plug socket to ON (“I” indicates
“ON”, “O” indicates “OFF”)

  1. Software
  2. Getting Ready
    ● For TissueGrinder Software to run successfully, the computer
    must meet the following requirements
    Windows 10
Microsoft .NET Framework 4.7.1 or higher
    Administrator privileges

● Plug in USB and power supply and switch to the ON Position

● Installing TIGR Software from the delivered USB Stick

● An "install wizard" guides you through the installation of
TIGR - just follow the prompts

  1. Starting the TissueGrinder
    ● The TIGR is used as a benchtop instrument. A number of
pre-defined protocols offer a variety of optimized protocols for tissue
dissociation, which can be selected in the Software

● The instrument allows creation and use of customized protocols for
specific applications. Standard protocols are accessible and can be
modified here: C:\ProgramData\FFX\TissueGrinder\Protocols

● Original TissueGrinder Grinding Consumables are required for
processing the samples
● To start the instrument:

Ensure that the device is plugged in USB and power supply before setting the switch to the ON position

Open TIGR Software by clicking its desktop icon or by using the Windows Start button and selecting: Programs > TIGR
The instrument starts within a few seconds and displays the TTIGR Slot overview. Illumination of the white LED indicates that the instrument is ready for operation. If an error message “Failed to initilalize one or more grinder” appears at the initial start, see section 6 Troubleshooting in the manual

  1. Operating the TissueGrinder

Note: Do not switch off the instrument while running a protocol to avoid severe damage of the instrument!

TIGR Workflow
Note: The following instructions assume that the instrument was successfully installed and initiated as outlined in TIGR manual or Quick Installation Guide.

Sample sizes range from and 20 to
400 mg of tissue and 300μL to
maximal 800μL total volume
To process samples, transfer an appropriate
amount of tissue and buffer or other
appropriate fluid into TIGR tube
Tightly close the tub beyond
the first resistance
Please make sure that the tube
is inserted and removed in an
upright position without tilting
Larger pieces of tissue
require manual cutting
into small pieces
On completion of the dissociation protocol, the
status of the LED will change from blue to green.
Remove the tube and put in the centrifuge.
Name the experiment and select the
required experiment and protocol.
Press Start to commence the dissociation.

  1. Sample Preparation
  2. Tissue Dissociation
  3. Centrifugation

Cell Strainers are available with three mesh
sizes: 40μm, 70μm and 100μm.

The tubes are for single use only.

Up to four samples can be run
simultaneously using the same or different
protocols.

Note: Do not switch off the instrument while running a protocol to avoid severe damage of the instrument!
